雷蒙特红豆草下胚轴愈伤组织原生质体再生植株

【摘要】 雷蒙特红豆草10d龄无菌苗的下胚轴切段在附加2,4-D和KT各1mg/L的MS培养基上诱导愈伤组织。取培养3周后形成的愈伤组织进行酶解可获得大量原生质体。原生质体在修改的KM8P液体培养基中培养,第10d开始分裂,2周后形成小细胞团,4周时发育成大细胞团。第7周将小愈伤组织转于含0.5%琼指糖的固体培养基上,它们可以旺盛生长,成为有根、芽分化的胚性愈伤组织,转于分化培养基上发育成苗和小植株。无根苗转于附加有NAA0.2mg/L、BA0.05mg/L、蔗糖2%的MS培养基中生根。

【Abstract】 Calli were from hypocotyl explants cultured on medium containing 2,4-D 1 mg/L, KT 1 mg/L. The protoplasts were isolated from primary calli and cultured in modified liquid KM8P medium. The first division of the regenerated cell occured within 10 days.Small cell clusters were formed and then developed into the large cell colonies 2 and 4 weeks after the culture , respectively. When transferred to 0. 5% agarose solidified media , protocallus proliferated and turned into embryogenic calli with differentiated roots and buds.Plantlets and multiple shoots developing on calli were subcultured MS medium of BA 0. 5~1mg/L and NAA 0. l ~0. 5 mg/L. Roots were induced from shoots on MS medium containing NAA 0. 2 mg/L. BA 0. 05 mg/L and sucrose 20%.
