金荞麦根体外培养人肿瘤细胞的抗癌研究
Antlcancer Effects of Fagopyrum Cymosum(Trev) Root Meisn on Human Tumor Cells in Vitro
【摘要】 利用直接杀伤细胞法,集落培养抑制法及DNA前体物质参入法研究金荞麦根对体外培养的多种人癌细胞的抗癌作用。结果:此药在1g·L-1时对多种人癌细胞的杀伤率均超过一个对数杀灭,浓度降低至0.125g·L-1的杀伤率仍接近一个对数杀灭达74.3%~92.l%。金荞麦根中的提取物具有明显抗癌作用,其浓度为0.1,0.05g·L-1对多种癌细胞的集落抑制率达100%,浓度为0.0125g·L-1时抑制率达75.l%~89.2%。再用3H-TdR标记法观察发现:金E能明显抑制癌细胞内的核酸代谢,其抑制作用与同浓度的阳性对照氟尿嘧啶近似。
【Abstract】 Fagopyrum Cymosum(Trev) root Meisn is a Chinese herb belonging to Fagopyrum,Hall. It has markedly amticancer effects. In this paper We report that, the anticancer mecha-nism of Fagopyrum Cymosum(Trev) root Meisn(FCM) on several lines of human can-cer cells. Experiments revealed that FCM had a remarkable killing effect on several lines ofhuman cancer cells in vitro。 The killing rates were greater than one log-kill when the tumorcells were treated with the drug in high concentration。 When the tumor cells were treated withthe drug(12.5 mg·L-1), the killing rates were 74.3%-92.1%。 Polygonum Cymosum(Trev)E(PC,E)is a highly active anticancer compound which was extracted from FCM。The colony formation experiment showed that the inhibition rates of colony formation were100% when the tumor cells were treated with C,Ein high concentration, when the tumor cellswere treated with PC。 E in low concentration ( l 2. 5mg·L-1), the inhibiting rates were80 %-100 %。 In 3H-TdR incorporation experiment revealed PC。 E had markedly inhibitedthe effects on biosythesis of DNA in the tumor cell5, its inhibition rate was greater than thatof 5-fluorouracil, at l0mg·L-1。
