应用高效液相色谱技术(HPLC)测定血清腺苷脱氨酶活性
The Application of HPLC in Determining Serum Adenosine Deaminase Activity
【摘要】 应用HPLC法改进血清腺苷脱氨酶(AD)活性的测定,对该酶反应主要因素如血清浓度、底物特异性进行比较实验。并测定34例健康人血清AD活性,其正常活性值0.255±0.075,此法较传统测氨法具有较多优点,如准确性高、重复性好和操作简便。此外,还可适用于检测血清中尿酸含量变化,有助于诊断肾脏疾病和老年痛风症。
【Abstract】 Serum adenosine deaminase (AD) activity is considered an indicator in routine diagnosis of hepatic diseases. In this paper,HPLC is used to determine inosine instead of using the classical ammonia method.The basic experiment was designed to determine the retention time of standard samples, serum concentration and substrate specific property. 34 Healthy indivduals were tested and the normal AD value was found to be 0.255±0.075.The authors hold that HPLC has more advantages such as high accuracy, good reproducibility and easy operation, and can also be used to determine serum uric acid, which can help the diagnosis of renal diseases and gout in the aged.
