生物免疫制剂对大鼠枯否细胞释放过氧化氢的影响
EFFECTS OF BIOLOGICAL IMMUNE STIMULANTS ON H2O2 RELEASE OF RAT KUPFFER CELLS IN VITRO
【摘要】 从正常Wistar大鼠肝中分离出大量高纯度的枯否细胞(KC),体外培养并以干扰素(IFNα—2b)、卡介苗(BCG)或二者联用处理枯否细胞,用荧光法检测免疫制剂对枯否细胞释放H2O2的影响.结果显示,IFNα-2b或BCG均可增强枯否细胞释放H2O2量,分别使释放量提高约1.6倍和1倍;IFNα-2b和BCG协同作用的效果更强,使枯否细胞释放H2O2量提高2倍以上.文章讨论了免疫制剂增强枯否细胞的H2O2释放量与其提高KC抗肝癌作用的关系及相关机制.
【Abstract】 Large numbers of purified Kupffer cells (KC) were isolated from rat livers and treated respectively with IFNα- 2b, BCG and the combination of IFNα- 2b and BCG in vitro, with the method of fluorescence assay, the effects of immune stimulants on H2O2 release of KC were studied. The results showed that the volumes of H2O2 produced by KC after treatment with IFNα- 2b or BCG increased by 1 time or 1.6 times compared with those of controls. Under the effect of IFNα- 2b and BCG, the content of H2O2 increased by more than 2 times. The relationship between increase of H2O2 released by KC and their enhancement of anti-hepatoma functions is discussed in the paper.
