【摘要】 用４种辣根过氧化物酶标记的凝集素，对牛精子表面糖复合物的分布进行了定位研究，并观察了固定和洗涤对糖复合物的影响。在直接涂片的精子上，麦芽凝集素、伴刀豆球蛋白Ａ、蓖麻凝集素Ⅰ和大豆凝集素均集中标记于顶体区，前三者在尾部亦有中等强度或弱标记。用成二醛固定对精子表面的凝集素标记无明显影响。但在洗涤后涂片的精子上，有些在顶体区部分或全部脱落一层膜样结构，从而导致了同一凝集素在不同的精子上表现出了不同的标记。更多还原

【Abstract】 Four HRP-labeled lectins of WGA,Con A,RCA I and SBA were used to study the localization of the glycoconjugates on the surface of bovine sperm and to elucidate the effects of fixation and washing on glycoconjugates,Marked differences were observed between the sperm samples directly smeared and three times-washed before smear,but fixation with glutaraldehyde showed no apparent effect.On the sperms directly smeared,1.WGA binding was very strong on the acrosomal region,weak on the postacrosomal region and moderate on the tail;2.Con A binding was strong on the acrosomal region and moderate on the tail;3.Both RCA I and SBA mainly bound on the apical portion of the acrosomalr region with moderate and strong intensity,respectively; After the sperms were washed three times with PBS and centrifugation,the binding pattern of lectins was apparently changed.In some sperms a piece of membrane-like structure on the acrosomal region partially or totally detached.At the acrosomal region where membranelike structure detached,Con A and RCA I remained moderate and weak bindings,respectively,however WGA and SBA showed negative binding in spite of a bandlike binding of WGA on the equatoral region,The results of the present study indicate that 1.glycoconjugates were plentiful and regionalized with a dominant localization on the acrosomal region,and 2.washing could alter the localization of glycoconjugates on some sperms.更多还原
