北京鸭淋巴细胞中间纤维-核纤维-核内骨架体系与淋巴细胞分化关系的形态学研究
Intermediate Filament -Nuclear Lamina-Nuclear Skeleton Systern in Peking Duck Lymphocytes from Bursa Fabricius with Special Reference to Lymphocyte Differentiation_Morphological Study
【摘要】 用DGD包埋超薄切片的电镜观察表明,北京鸭腔上囊淋巴细胞的分化过程中,中间纤维-核纤层-核内骨架体系变化明显。淋巴母细胞的中间纤维呈致密的网络状,核纤层较薄,核内骨架纤维网络稀疏。中淋巴细胞和小淋巴细胞的中间纤维网络变得稀疏,核纤层增厚,核内骨架呈现致密的纤维网络状结构,其上附着许多颗粒样物质,抗波形蛋白抗体和抗核纤层蛋白抗体的间接免疫荧光染色同样表明,淋巴母细胞胞质的波形纤维呈网络状,核纤层呈弱阳性反应;中淋巴细胞和小淋巴细胞的胞质波形纤维网络结构不明显,核纤层则呈强阳性反应。结果提示,淋巴细胞中间纤维-核纤层-核内骨架体系的变化与淋巴细胞增殖和分化过程密切相关。
【Abstract】 By using selective extraction method and emteddment-free ultrathin section eltctron mi-croscopy , Peking duck lymphocytes isolated from bursa Fabricius of different divelopment stages were studied It was shown that lymphoblasts had thicker interconnecting nrtwork of intermedi-ate filaments in cytoplasm and thinner nuclear lamina as well as thinner network of nuclear skele-ton as compared with medium lymphocytes and small lymphocytes. Indirect immunofluorescence method for demonstration of vimentin and lamins was also employed. The lymphoblasts were positive for vimentin, and their vimentin filaments organized into network. These cells were weakly positive for lamins. and lamin fluorescence distributed at the periphery of nuclei. In the medium lymphocytes and small lymphocytes, positive vimentin staining were seen while vi-mentin network was not clear. These cells showed brighter fluorescerce for lamins. The aboveresults suggested that the changes of inttrmediate filament-nuclear lamina-nuclear skeleton sys-tem in Peking duck lymphocytes correlate closely with difftrentiation of lymphocytes.
