杂交瘤细胞培养保护剂的研究
Studies of Protective Agents for Hybridoma Cell Culture
【摘要】 利用鼠鼠杂交瘤2F_7细胞(分泌IgG_2单抗),研究了普兰卢内克(I)、聚乙二醇(II)、甲基纤维素(III)、葡聚糖(IV)、聚醚多元醇(V)与杂交瘤细胞的生物相容性、添加限制浓度以及高速搅拌下添加剂对杂交瘤细胞的保护作用。实验结果表明,0.05%~0.1%的I、0.04%~0.20%II、0.10%~0.20%的III能较好地保护杂交瘤细胞;IV会抑制细胞生长;v会分解细胞。在1.5L生物反应器中培养,添加0.10%I于培养基,搅拌转速为70r/min时,细胞能正常生长。
【Abstract】 The effects of different protective agents such as pluronic F-68,polyethyleneglycOl (PEG),methylcellulose(MC),dextran, on hybridoma 2 F7 cell culture which secretesmonoolonal antibody against small cell lung cancer were studied. The potential toxicity,restricting adding concentration, protective effeccs at high agitation speed were investigated. It is shown that 0.05%~0.1%pluronic F-68,0.04%~0.2%PEG,0.1~0.2%MC canprotect hybrdoma cell from shear stress at high agitation-speed,dextran inhibits cell growth. In 1.5 L celligen bioreactor, adding 0.1% pluronic F-68 in medium,at 70 r/min of agitatingspeed,the hybridoma cells can normally grow.
