引流熊胆对培养鼠肝枯否细胞的免疫活性影响及电镜观察

【摘要】 将Wistar系大白鼠(乳鼠)肝枯否细胞体外培养4—8天,取细胞生长良好的培养瓶,分为正常组、损伤组、实验组和阳性对照组,利用特异抗体的介导,检测枯否细胞免疫活性并进行电镜观察。结果,正常组与损伤组、实验组与损伤组之间差异显著(P<0.01),而正常组与实验组无明显差异,接近阳性对照组。电镜下,正常组枯否细胞表面有许多突起,胞质内含吞噬的绵羊红细胞(SRBC),溶酶体丰富,空泡多,线粒体、粗面内质网较发达,尚可见髓样结构,膜被囊泡与吞噬体及溶噬体;损伤组枯否细胞的细胞器损伤不很明显,有的细胞线粒体肿胀,局部细胞膜三层结构模糊不清,吞噬的SRBC数目比正常组明显减少或无SRBC;而实验组枯否细胞线粒体清晰,吞噬的SRBC明显增加,与正常组及阳性对照组相似。四组吞噬活力比较:阳性对照组>正常组>实验组>损伤组。

【Abstract】 Having cultured the hepatic kupffer cell of wistar rat (infantile rat) in vitro for 4 ̄8 days, we selected the culture bottles in which .the cells were growing better, and divided them into four groups: normal group, injuried group, experimental group and positive control group. With the intervention of specific antibody, several immunologic functions of kupffer cells were investigated and its appearance under the electron microscope was observed. The results revealed that the garland forming rate and the percentage of phagocytosis of kupffer cell showed significantly difference between the normal and injuried group, and between the experimental and injuried group, (P<0.01), but no difference was found between the normal and experimental ones. Under the electron microscope, it was showed that in the normal group, there were many processes on the kupffer cells surface, phagocytized sheep red blood cell (SRBC), and large amount of lysosomes as well as mitochondria, relatively abundant RER in the cytoplasma and it was also showed myelinic structure, membrane--coating vesicle, phagosome and so forth. However in the injuried group the injury of kupffer cell was not obvious. The mitochondria was turgencent in some of the cells, the structure of the cell membrane was blurred partly, but the number of phagocytized SRBC was decreased significantly. The experimental group was similar to the normal and the positive control ones. The mitochondria was clear and the number of phagocytized SRBC was increased obviously. The comparison of phagocytic power showed as follows: the positive control group>the normal group>the experimental group>the injuried group.
