离体接种鉴定水稻品种的抗瘟性
In Vitro Inoculation Technique for Identification of Rice Blast Resistance
【摘要】 研究了离体稻叶和稻穗接种鉴定水稻品种抗瘟性的方法.离体稻叶接种,采用水稻田、五叶期顶端第一片完全展开叶,用100×10-6-200×10-6苯骈味唑溶液作保绿剂,以20×10-6-30×104个/mL的稻瘟病孢子悬浮液喷雾接种.离体稻穗接种,剪取稻穗,保留穗颈下一茎节和剑叶,插在培养液中,在室内用浓度为10×10-6-20×104个/mL的孢子悬浮液接种.结果表明,不管是叶瘟还是穗瘟,离体接种与非离体接种的鉴定效果基本一致.因此,离体接种方法可以广泛地运用于各种抗瘟性鉴定.
【Abstract】 The inoculation technique for identification on rice blast resistance in vitro was studied. In vitro leaf blast inoculation was carried out by spraying 20×104-30×104 conidia/ mL suspended onto the segments at stage of the 4-5 fully expanded uppermost leaf which were kept green using 100×10-6-200 ×10-6benzimidazole solution in culture dish. In vitro panicle blast inoculation was carried out by inoculatig 10×104-20 ×104 conidia / mL suspended to the rice panicles which were resided the uppermost stem and leaf and inserted in culture. The results showed the infection effects both on leaf blast and panicle in vitro were consistant with inoculation in vivo. Thus, in vitro inoculation technique could be used blast resistant identification extensively.
