圆周免疫扩散法检测人血浆纤维蛋白原的研究

【摘要】 本文介绍一种特异、简便测定人血浆纤维蛋白原(Fg)的圆周免疫扩散法。本法采用纯化的人Fg免疫山羊制备单价特异的抗血清、抗体凝胶板每孔点样5ul,25℃-37℃扩散48小时测量免疫反应沉淀环直径。标准曲线工作范围为1.5—6.0g/L。最小检测量为1.26Mg/孔。板内及板间变异系数分别为1.02—2.95%及1.17—3.33%。样品回收率为95.3—103.7%。用本法测定成都地区100例健康成人和33例高甘油三酯血症患者血浆Fg的含量分别为3.25—0.472g/L和3.62—0.67g/L(M±SD)。与免疫浊度测定法比较其相关系数r为0.9259,p<0.0005。

【Abstract】 A radial immunodif fusion assay ( RID ) for quantitation of human plasma fibrinogen was developed. Antisera to human fibrinogen were raised in goats with purified fibrinogen The antibody-gel plate was prepared with 12g/L agarose. 5μl of each sample or standard plasma was added in a well. Diffusion was performed at 25℃-37℃ for 48h. The standard curve with a working range of 1.5-6.0g/L was plotted. The minimum measurable concentration of fibrinogen was 1.25μg. The coefficients of variation of the reproducibility ot intra-assay and intra-assay were 1.02-2.95% and 1.17-3.33% respectively. The recovery of the assay was 95.3-103.7%. The concentration of fibrinogen for healthy adults and palients with hypertriglyceride in Cheng Du area were 3. 25±0.472g/L (M±SD, n = 100 ) and 3.62±0.67g/L ( n = 33 ) respectively. The cor rrelation to immunoturbjditric assay ( ITA ) was 0.9259 P<00005.
