鼠疫菌pH6抗原的电泳图型识别及其体外表达条件

【摘要】 pH6抗原是鼠疫菌毒力的重要组成部分,研究结果显示,这种抗原很可能与腺鼠疫的发病机制有关。为提供国内急需的pH6抗原纯品,研究和观察了该抗原的体外适宜表达条件。结果表明,在四种常规培基中,pH6抗原均可表达,但在LB培基中,该抗原的表达量明显高于其他培基。此外,还进行了两种pH及两种温度条件下该抗原表达量的比较,如预期的那样,该抗原仅在酸性和37℃条件下最大限度表达。根据该抗原不同时PH和温度条件下十二烷基磺酸钠—聚丙烯酰胺凝胶电泳(SDS—pAGE)图型的比较、参照已单体分子量,初步识别了该抗原的电泳图型,为今后进一步提取、纯化这种抗原成分奠定了基础。

【Abstract】 H6 antigen is important composition associated with virulence of Yersinia pestis。The evidences supported that the antigen probably involved in the pathogenesis of bubonic plague。In order to supply the purer antigen,the suitable conditions were investigated in our experiment.The resuIts showed that the antigen could beexpressed in four routine mediums and the yield of this antigen in LB edium was prominently higher than inother mediurns。On the other hand,comparison of pH6 antigen expression quanlity was carried Out under twokinds of pH and temperattire respectively。As expected,it was only under the condition of acidity and 37℃that the antigen was expressed as possible as it could be。According to the contrast of SDS- pAGE patternsof pH6 antigen under different pH and temperature,as well as consulting the known monomer molecularweight,a electrophoretic pattern of the antigen has been discerned initially。We wish that the study could con-tribute to the extrection and purification of this composition from agent。
