红缘层孔菌多糖FP2的结构与体外抗肿瘤作用的研究

【摘要】 从红缘层孔菌子实体中分离纯化-葡聚糖,MW约为214Kd,红外光谱证明其含β型糖普键,比旋值[α]15为+49°,ηsp为0.22,凯氏定氮及紫外光谱分析证明FP为蛋白多糖。FP与刚果红络合及NaOH浓度对其粘性行为的影响均显示β(1→3)GLc为主链的分子特性。高碘酸氧化、Smith降解、甲基化分析证实FP确以β(1→3)GLc为主链,其6位上带有分支点,平均每3个糖残基带有1个支链。β-消除反应表明FP中的蛋白质是以Thr-0-糖苷键及Ser-0-糖苷键相联。FP于体外对S-180肿瘤细胞无抑制作用。

【Abstract】 A glucan has been isolated from Pinioola Fomes fruit bodies. Its molecular weight was about 214Kd. [α] = 49°, η(SP)= 0. 22, FP2 contain 1. 8% protein. Complexformation with Congo Red and effects of concentrations of NaOH on indicated character of main chain composed of β (1→3) GLc. The structure of FP2 was determined by periodate oxidation, Smith degration, methylation, I. R etc. The main chain of FP2 is composed of β(1→3) linked GLc, Single β (1→6) linked GLc attached, on average, to every three sugar residue of the main chain. The polysacchrides combine protein by forming o-glycosidic bonds through Thr and Ser, FP2 has been examined by β-elimation. FP2 was not found the antitumor activity on the Sarcoma 180 cells.
