~3H-甘露聚糖在小鼠体内吸收与代谢的初步研究
Studies on Absorption of Large Molecular Mannan After Oral
【摘要】 给小鼠灌胃~3H-甘露聚糖(SP_1)后,30min即可从血液中测到放射性,60min血中放射性达高峰;于不同时间对各组织放射性的定量测定及小鼠整体切片的放射自显影结果均证明,~3H-甘露聚糖经口服可被机体吸收。进入小鼠体内的放射性物质主要从粪与尿中排泄,从胆汁排泄较少,给药后48h区间内,从粪、尿、胆汁中累积排出的,总放射性分别占给药剂量的42%、29%、5.4%。经Sepharose-DL-4B柱层析鉴定,经口服吸收入血的SP_1分子量不变。给药后1h,血清中的SP_1约占给药剂量的11%。经纸层析对代谢物鉴定,粪中有22%为原型药物,尿中未检出原型药。
【Abstract】 Radioactivity(4.6 ±4.3dpmx10 ̄5/mL)could detected from the blood 30 min after ig administration, and reached its peak(31±9.8dpmx105/mL)at 1h,absorption of H-Mannan from gastrointestinal tract in mice was rapid. Study on tissue distribution showed that radionctivity was found to be highest in the liver(28±6.7dpmx104/100mg)and kidney(24±4. 5dpmx104/100mg), 2h after ig administration.Result of who.le body autoradiography of mice showed that H3-Mannan could be absorbed by body. Radioactivity mainly excreted from feces and urine. Cumulative radioactivity excreted in feces,urine and anobile was 42%, 29%,5.4%, respectively, within 48h,after ig admi-nistration,pharmacokinetic behavior of H3-mannan after ig was similar to that of after injection. Determination of mannana SP1 in serum of mice by chromattography on Separose-CL 4B column showed SP1 absorpted from gastrointestinal tract, its molecular weight is not changes after ig 1h, SP1 of serum was 11% of the dose per os. Metabolite in feces detected by P.C.SP1 was 22%.
