血清中GOT和GPT活性连续测定的双酶膜电极的研制

【摘要】 介绍了用双酶电极对两种转氨酶GOT和GPT进行检测的原理和方法,研制出一种可用于临床上分析GOT和GPT的酶传感器,对两种转氨酶电极最佳测试条件:25℃,pH 7.3,电极响应时间:GOT为3分钟、GPT为5分钟、固定化的酶电极工作寿命14天,测定样品400次,酶膜在4℃冰箱中保存6个月,其中POP为原活性80%,OAC为原活性85%,该法与临床用自动化分析仪比较P>0.05两者符合良好。

【Abstract】 The principles of determination of GOT and GPT using an immobilized bienzyme-Poly membrane electrode is described in this paper. The biosensor is used in the sequential determination of GOT and GPT in Clinic. The optimal analytical parameters of GOT and GPT: temperature 25℃ . pH7,3. Response time of GOT is 3 min. Response time of GPT is 5min.The apparent enzyme activity of bienzyme membrane electrode retained 70% of the initial activity after 400 assays over 14 days. Moreover the immobilized eneyme retuned 80% for pop and 85% for OAC their initial activity after storage at 4℃ for 6 months.We tested the electrod and biochemical autoanalyser the results show good agreement.
