激光光散射及荧光光谱法研究金属离子对牛磺胆酸钠胶团性质的影响
Studies on the Influence of Metal Ions on the Colloid Properties of Sodium Taurocholate by Laser Scattering and Fluorescence Spectroscopy
【摘要】 采用激光光散射的方法,研究了Ca ̄(2+)、La ̄(3+)、Eu ̄(3+)存在下,NaTC胶团及其与胆固醇、卵磷脂形成的混合胶团平均流体力学半径变化,发现金属离子能使胶团长大,并对胶团的结构及对胆固醇的增溶性有影响。以芘为荧光探针,研究了Ca ̄(2+)、La ̄(3+)离子对胶团内部微极性的影响,及通过Eu ̄(3+)的荧光光谱表征其配位场特征,证明金属离子与胆盐胶团间相互作用发生在胶团的表面。
【Abstract】 In this report,the influence of metal ions(Ca ̄(2+),La ̄(3+),Eu ̄(3+))on the mean hydrodynamic radii of taurocholate micelles or its mixed micelles with cholesterol and lecithin was studied by laser scattering method,and its mechanism was investi-gated by fluorescent labeling and fluorescence spectra. The results show that adding of metal ions makes micelles grow and the effect is decided by both metal ions and micelles. For metal ions,the effect is La ̄(3+)>Eu ̄(3+)>Ca ̄(2+).The existence of Ca ̄(2+),as well as lecithin,increases the solubility of cholesterol by taurocholate micelles.For mixed micelles,the mean hydrodynamic radii increase with the addition of Ca ̄(2+)which may cause the change of micellar structure and shape. The addition of HCl causes the Rh of micelles increase in the system without CaCl2 and decrease in that with CaCl2.Fluorescence results show that the coexistence of metal ions and mi-celles does not affect micellar micropolarity or coordination field of metal ions,which indicates that the interaction between metal ions and taurocholate micelles may occur at the surface of micelles.
