热毒清防治内毒素性DIC(续)——保护肝微粒体、钙稳态和抗自由基的实验研究
Prophylactico-therapeutic effect of Re Du Qing on endotoxin-induced DIC: Protecting hepatocytic microsomes, aintaining Ca++ homeostasis and antagonizing oxygen free radicals
【摘要】 采用内毒素制作家兔DIC模型,观察到模型组动物肝微粒体细胞色素P-450酶含量和苯胺羟化酶活性明显降低;肝线粒体膜脂微粘度明显增高,膜流动性和Ca2+-Mg2+-ATP酶、Ca2+-ATP酶明显降低;肝组织等脂质过氧化物和黄嘌呤氧化酶显著升高,超氧化物歧化酶和谷胱甘肽过氧化物酶显著降低(P<0.05、P<0.01)。而热毒清治疗组上述变化均不明显而接近正常对照组。提示:在内毒素DIC时,肝微粒体受损,钙稳态失调,自由基堆积过多;而热毒清则具有保护肝微粒体,维护钙稳态和防止脂质过氧化作用。
【Abstract】 In this study, DIC model of rabbits was made by injecting endotoxin, The results in model group showed that cyto. P-450 countents and aniline hydro- xylase activity in hepatocytic microsomes were de- creased remarkably, the microviscosity of mitochondria membrane was increased while the fluid- ity of the membrane, and Ca2+, Mg2+, ATPase as well as Ca2+, ATPase activity of mitochondria were de- creased conspicuously. Furthermore, the levels of lipidperoxide(LPO) and xanthine oxidase(XOD) in hepatic tissue were raised whereas superoxide dismutase (SOD) and glutamine peroxidase (GSH-Px) were reduced distinctly in comparison with the normal control group (all P< 0.01). The parameters in rabbits treated with Re Du Qing (RDQ group) were significantly different from model group and similar to normal control group. The results re- vealed that endotoxin may causehepatocytic microsomal damage, calcium homeostasis disturbance and excessive accumulation of oxygen free radicals, on the other hand, Re Du Qing possesses the effects on protecting hepatocytic microsomes, maintaining Ca2+ homeostasis and antagonizing oxygen free radi- cals.
