蔗糖曲拉通X—100裂解G~+、G~-菌质粒提取方法
A Method of Plaunid DNA Preparation by Sucrose-TritonX-100 Detergent from Gram-positive and Gram-Negative Bacteria
【摘要】 本文报告一种质粒DNA的提取方法,该法首先用100%蔗糖处理细菌,然后用tritonX—100—脱氧胆酸盐裂解,无需溶菌酶也不需昂贵的仪器。结果表明,该法对标准质粒菌E、coliV517的质粒检出显示很好的重复性。对金黄色葡萄球菌、铜绿假单胞菌、痢疾杆菌的质粒检测结果分别为88.5%(23/26)、67.4%(31/46)、100%(7/7)、对金黄色葡萄球菌的大、小质粒都能检出。本方法简便经济,是兼适用G~+和G~-菌的质粒分析方法。特别适于流行病学调查。
【Abstract】 The article reports a method of plasmid DNA preparation from bacteria. In this method,firstly,bacteria is treated with containing 75% sucrose and then is lysed by the lysis mixture, containing triton x-100 and sodium deoxycbolate. It does not require costly chemicals e、g、lysoslaphin, lysozyme, as well as expensive equipment. The results indicated that the reproducibility of plasmid DNA preparation was very satisfactory for standard strain of E. coli V517 and 88.5%(23/26) of staphlococcus aureus, 67.4%(31/46)of Pseudomonas aeruginosa and 100%(7/7)shigeliae were found plasmid positive. It was simple and inexpensive method of plasmid from both gram-positive and gram-negative bacteria. It was suitable especially for the epidemioiogical investigations of bacteria.
