【摘要】 用~3H-半乳糖掺入与软琼脂克隆培养相结合测CSA的方法,初步建立起对巨噬细胞产CSFs有促进作用的药物筛选模型。在用此模型对茯苓素的筛选中,我们发现茯苓素有效部分10150及从10150中分离出的单一组分10152在体外能增加LPS活化的小鼠腹腔巨噬细胞产CSFs的能力,且10152的增强作用有剂量依赖性关系。在体内实验中,腹腔注射10150能使小鼠血清中CSA增高,外周血白细胞水平上升。另外,用腹腔注射环磷酰胺造成的小鼠外周血白细胞减少模型,我们发现先期腹腔注射10150能预防由环磷酰胺引起的白细胞减少(白细胞下降幅度由56％减少到37％),而在每次腹腔注射环磷酰胺(25mg/(kg·d),连续四天)后1小时再腹腔注射10150(25mg/(kg·d),连续四天),能使白细胞下降幅度明显减少(白细胞下降幅度由53％减少到27％),且恢复时间提早2天,效果强于升白药阿魏酸钠。更多还原

【Abstract】 By taking colony stimulating activity (CSA) as the selective index, combining the ~3H-galactose incorporation assay and the sern-isolid colony test, we set up a drug screening model to detect active components which can improve the production of CSFs by macrophages, lymphocytes and other cells.By using this model, we have found an effective portion 10150 and an effective single component 10152 from Poria-tin. In vitro, 10150 and 10152 both improved the production of CSFs by LPS stimulated murine peritoneal macrophages and the activity of 10152 is dose-dependent. In vivo, we have also found that 10150 enhanced both CSA in the murine serum and WBC in peripheral blood after intraperi-toneal administration. In the model of murine leukocytopenia caused by administration of Cyclophosphamidum (CY), we have found pre-treatme-nt with 10150 can prevent leukocytopenia. The administration of 10150 (25mg/(kg·d)) followed the treatment with CY (25mg/(kg·d)) for four consecutive days can reduce the decreased range of WBC (the decreased range was reduced from 53% to 27%) and shortened the period of leukocytopenia for 2 days. And the efficacy of 10150 is better than Sodium Fe-rulate.更多还原
