柚汁胞愈伤组织的诱导及耐盐性的研究
Studies on callus in vitro of pomelo juice vesicles and its tolerance of salt.
【摘要】 本试验以柚汁胞为培养物,脱分化产生愈伤组织。研究了汁胞愈伤组织布不同NaCl浓度处理下矿质营养元素的变化。试验表明:在MT附加2,4-K 0.5+BA 1.0.培养基中,柚汁胞愈伤率达98.5%。愈伤组织几乎都是从汁胞柄上产生。在NaCl盐逆境下,愈伤组织生长受抑制。在NaCI 0%~0.5%范围内愈伤组织能够生长,在1.0%~2.0%范围内,生长量不到对照的10%。则定的13种矿质元素中,愈伤组织内Na~+含量为外源Na~+浓度的2.3~4.6倍。而P,K,Ca与Na~+含量成线性相关。
【Abstract】 Calli of pomelo [Citrus grand he (L. ) Osbeck] juice vesicle had been induced in vitro. The changes of mineral elements in concentration in calli with the difference NaCl concentration had been studied. Exogenous hormone combination was 2,4-D 0. 5, BA 1. 0. The percentage of induced calli was 98. 5%. Almost all of the calli initiated from the matural vesicle stalks instead of sacs. The growth of the calli was permitted within the range of 0%-0. 5% NaCl, hut was inhibited and less than 1% in contrast with control when the medium containing NaCl was more than 1%. The different contents of 13 different mineral elements in the calli were determined under salt stress of NaCl. The concentration of Na ’ in caili was 2-4. 5 times that of exogenus Na-. K - / Na + ratio decreased rapidly when the concentration of exogenus NaCl increased. P. K. Ca Had linear correlation with the foments of Na+ in the calli.
