用脱乙酰几丁质制备固定化青霉素酰化酶

【摘要】 以青霉素酰化酶基因工程菌为酶源,选用脱乙酰几丁质为载体,试用了4种固定化方法,从4个方面(pH、温度、盐浓度和结合酶量)摸索了固定化条件。结果在适宜条件下获得比活900U/g(干)(NIPAB法)的固定化青霉素酰化酶,回收率可达56%。并测定其酶学性质。

【Abstract】 Chitosan has been used as the carrier to immobilize the penicillinacylase(PA) which was extracted from E.coli 108/pPAHD1 constructed by DNArecombinant technique.Three different methods and 4 conditions(pH,tempe-rature,NaCl concentration and quantity of PA to be immobilized) have been con-sidered.Under the suitable conditions,the activity of immobilized PA was ashigh as 900 U/g(dry),and the activity retention was around 56%.The propertiesof the immobilized enzyme were determined.
