HPLC测定初发甲亢病人体内丙基硫氧嘧啶药代动力学参数
Serum propylthiouracil concentration and its pharmacokinetic parameters in hyperthroid patients: determination byHPLC
【摘要】 用高效液相色谱法测定丙基硫氧嘧啶血药浓度。以咖啡因作内标,样品在酸性条件下采用氯仿—异丙醇混合溶剂进行抽提,于37℃水浴上吹干,残留物用甲醇溶解进行色谱分析。分析柱(250mm×4.6mm,ID)内填国产YWG-C18固定相。流动相为含8%乙腈的O.025mol/L磷酸二氢钾水溶液。检测波长为272nm。用丙基硫氧嘧啶与内标的峰高比进行定量,其线性范围在0.1~4μg,(γ=0.9982),最低检测限可达0.05μg。本文还报道了6例初发甲亢病人体内丙基硫氧嘧啶药代动力学参数。
【Abstract】 A highly specific and sensitive method us-ing HPLC for the determination of propylth-iouracil(PTU)in serum was established. Thecalibration curve was linear for the concentra-tion from 0.1~8μg/ml, r=0.9982. The rala-tive recovery was 95% to 101%, and its detec-taple serum concentration was 0.05μg/ml.PTU pharmacokinetic was studied after singleoral administration of 200 mg in 6 hyperthy-roid patients. Serum concentration-time datawere fit to one-compartment model. The meanpharmacokinetic parameters of PTU obtainedwere as follows: Ka=4.53h, K=0.947h, T1/2=0.792h, Tp=0.68h and Cp=6.04μg/ml.The advantages of this method are discussed.
