HPLC程序控制波长自动变换方法测定体内氮烯乙茶及其代谢产物

【摘要】 高效液相色谱微机程控自动变换波长的方法,使生物样品中的氮烯乙茶及其代谢产物可同时在各自的最大吸收波长下得以测定,保证了各组分的最大检测灵敏度。观察了小鼠血中该药及代谢产物的浓度变化。

【Abstract】 To determine etheofazine(λmax=340nm)and its metabolite(λmax=280nm)simultane-ously, a method of HPLC with microprocessorcontrolled auto-adjustment wavelength wasdeveloped. Every component could be deter-mined at its optimum wavelength by the con-trol of microprocessor. The method was par-ticularly advantageous for the determination ofa few components with individual λmax in thesame sample. The range of the linear relation-ship between peak area and concentration was0.2~15μg/ml. The limit of detection was0.1μg/ml, recovery was 87.2% with RSD=5.1% for etheofazine; 0.15μg/ml, 90.7%, and7.5% for metabolite.
