分泌抗赭曲霉素A单克隆抗体杂交瘤细胞株的建立
Establishment of a hybridoma cell line secreting monoclonal antibody against ochracin A
【摘要】 用赭曲霉素A(OA)-BSA合成抗原免疫Balb/c小鼠,取免疫鼠脾细胞与sp2/0细胞融合。通过克隆和ELISA法筛选,建立三株分泌抗OA单克隆抗体杂交瘤细胞株(3C12、1D12和2G7)。用间接ELISA法测定,细胞上清液抗体效价为128χ(3C12)和64χ(1D12和2G7),腹水抗体效价为10-7(3C12、1D12)和10-6(2G7)。三株单抗(McAb)’均属IgG类,分泌抗体稳定。用竞争间接ELISA测定三株McAb对OA的敏感性为22.5~39.2ng/ml,与异种毒素黄曲霉毒素B1和ZEN毒素无交叉反应。
【Abstract】 Using antigen that synthesized with ochracin A-BSA immunized Ba1b/c mice, extracting the spleen cell of the mice, and SP2/0 cell fused. Through clone and ELISA screen, set up three hybridoma cell lines (3G2, 1D12 and 2G7) of secretion against OA monoclonal antibody. Antibody valence of cell supernatant fluid, and ascites were 128, (3G2) 64(1D12, 2G7), and 10-7(3C12), 10-6(2G7) by ELISA respectively. Three strains of McAb belong to IgG. It secreted stable antibody. The sensitivity, of the three strains of McAb to OA was 22.5-39.2ng/ml by competitive indirect ELISA. It’s no cross reaction with aflatoxin B1 and ZEN toxin.
