柑桔耐盐系的离体诱发与原生质体植株再生
In Vitro Induction and Protoplast Plant Regeneration from NaCl-Tolerant Lines in Citrus
【摘要】 本研究用γ射线、EMS诱变,NaCl直接选择10代,获得‘锦橙’、‘桃叶橙’耐盐突变系。耐盐系在无盐胁迫下培养3代,经原生质体分离、培养后再回到盐胁迫下,存活率和生长都正常,这说明耐盐系是稳定的。耐盐系能积累更多的游离脯氨酸,维持较高水平的K~+,吸收较少的Cl~-和Na~+。这很可能是耐盐系耐盐的生理基础。从耐盐系珠心愈伤组织分离原生质体,培养2个月后形成胚状体,胚状体进一步萌发成耐盐原生质体植株。
【Abstract】 NaCl-tolerant lines of ’Jincheng’ and ’Taoyecheng’ (Citrus sinensis) were obtained by subjecting their nucellar calli to γ ray and EMS, and 10 generations of selection in vitro. Later tests confirmed that their tolerance was stable. The tolerant lines accumulated more proline, maintained higher level of K~+ , and absorbed less Cl~- and Na~+ than the original calli. The protoplasts from the tolerant lines developed embryoids after 2 months culture, and they germinated into plantlets. Besides, it was found that the mu-tagenic sensitity of cultured cells was affected by explant source and culture method as well as species (variety).
