内皮素-1对大鼠血管平滑肌的作用及川芎嗪的对抗效果

【摘要】 用离体大鼠的主动脉和培养的大鼠胸主动脉平滑肌细胞,研究了内皮素-1对主动脉的收缩作用及对主动脉平滑肌细胞增殖周期及能量代谢的影响,结果表明1.0~18.0nmol·L1的内皮素-1能引起浓度依赖性血管收缩,川芎嗪能有效对抗内皮素-1血管收缩作用.100~100.0 pmol·L1的内皮素-1能使S+G2+M期细胞的比例增加,具有促进血管平滑肌细胞增殖的作用;10.0 pmol·L1内皮素-1作用10 min可升高平滑肌细胞ATP含量,20 min之后ATP含量一直低于正常水平,内皮素1对平滑肌细胞ATP含量的影响呈双相作用,先升高后降低

【Abstract】 Endothelin I at concentrations of 1.0 18.0nmol·L-1 elicited a dose dependent contr-action of rat thoracic aorta in vitro.The maximum tension was larger than that induced by 30 mmol·L-1 KC1.The EC50 value of endothelin 1 was 3.7±s 0.7 nmol·L-1 The endothelin 1 induced contraction could be reversed by 5.7 23.0 nmol·L-1 tetramethylpyrazine hydrochloride.Under normal conditions,the percentage of the cultured vascular smooth muscle cells at S+G2+M phases was 27.5±s 1.6%.Endothelin 1 had a dose dependent proliferation promotion effect on the cultured vascular smooth muscle cells.At 24 h after exposure to endothelin 1,the percentage of S+G2+M phases cells increased to 40.7+s 2.2 and 45.6±s 1.3% at concentrations of 10.0 and100.0 pmol·L-1,respectively.The results show that endothelin 1 could promote the proliferation of the cultured smooth muscle cells.Using the biological luminescent method,the authors measured the changes of ATP content in the cultured smooth muscle cells.Endothelin 1 10.0 pmol·L-1 could induced a temporary increase(139±s 1%,P<0.01,n= 3)in ATP content of the cultured vascular smooth muscle cells as compared with the control group 10 min after the addition of endothelin 1,but at 20 min the ATP content was lower than the normal level.
