免疫毒素清除人骨髓中克隆化白血病细胞的研究——Ⅰ.抗CD7免疫毒素的制备及其细胞毒性质研究
ELIMINATION OF CLONOGENIC LEUKEMIC CELLS FROM HUMAN BONE MARROW WITH IMMUNOTOXINS I. THE PREPARATION AND CYTOTOXICITY OF ANTI-CD7 IMMUNOTOXINS
【摘要】 本实验采用优化的方法通过SPDP交联抗CD7单克隆抗体与蓖麻毒素制成免疫毒素。经MTT方法检测其对表达CD7抗原的Molt-4及HPB-ALL细胞系均有显著特异性杀伤作用。克隆杀伤实验结果表明,该免疫毒素在10-10mol/L浓度时达3个杀伤log(即清除99.9%的molt-4细胞),而在此浓度时,其只抑制21.6%的粗-单系造血祖细胞生长,从而初步证明其在自体骨髓移植中可用于清除骨髓中残留白血病细胞。
【Abstract】 The anti-CD7 immunotoxin was prepared by linking anti-CD7 McAb and Ricin with SPDP. The condition of reaction was improved to increase the production and cytotoxicity. The specific cytotoxicity of the immunotoxin was evaluated by MTT method. Its IC50 to Molt-4 and HPB-ALL cells, which express CD7 antigen on their surface, are 5×10-11mol/L and 10-9mol/ L, respectively. The result of cloning assay indicated that the immunotoxin could kill 3 log (99.9%) Molt-4 cells at the concentration of 10-10mol/L, but it inhibited only 21% of the growth of CFU-GM at that concentration, thus its preliminary efficacy in elimination of residual leukemic cells in ABMT is verified.
