自制简易胚胎冷冻器冷冻奶牛胚胎试验

【摘要】 用FSH+PGF2超排黑白花奶牛,以非手术方法采得7日龄早期胚胎。应用含10%甘油卵细胞培养液作为胚胎冷冻保护液,采用自制简易胚胎冷冻器冷冻,在-196℃液氮中保存。冻胚在35℃水浴中解冻,应用92.1g/L甘油+171.2g/L蔗糖PBSS,46g/L甘油+171.2g/L蔗糖PBSS和171.2g/L蔗糖PBSS三步脱去甘油,用含20%犊牛血清PBS将胚胎清洗2遍后装管移植。共解冻18枝冻胚,16枚可用。可用胚率为84.2%(16/18)。采用非手术方法移植16头受体牛,5头妊娠产犊,移植妊娠率为31.5%(5/16)。

【Abstract】 Donor Holsteins were superovulated with Fsh+PGF2, and flushed non-surgically at day 7. Cryoprotectant was cocyte culture medium containing 10% glycerol. Fmbryo freezing was accomplished by means of self-made freezier. The embryos were stored in liquid nitrogen at -196℃. Frozen embryos were thawed in a 35’C water bath, and the cryoprotectant was removed by three steps of 92. 19 g/L glycerol+ 171. 2 g/L sucrose in PBSS,46 g/L glycerol + 171. 2 g/L sucrose in PBSS. and 171. 2 g/L sucrose in PBSS. Embryos were washed twice in 20% PBS and transferred non-surgically into the uterus of recipients. Eighteen frozen embryos were thawed, sixteen of them could be used. The rate of available embryos was 84. 2% (16/18). Available embryos were transferred non -surgically to 16 recipients. Five of them be came pregnant. The transplant pregnancy rate was 31. 3% (5/16).
