【Abstract】 Embryos from the mature seeds ofHordeum vulgate L.cv.Zaoshu No.3 we-re allowed to germinate on MS mediumsupplemented with 1% sucrose.Variationof mitotic index(M.I.)in 0—3mm ba-sal region of first leaves was monitoredfor 49—72h,121—144h and 48,72,96,120,144h seedlings,and those in 0.5mmsuccessive segments between 0 and 10mmbasal region of first leaves for 120h seed-lings as well.For inducing callus,the3mm leaf segments from the base of 1stleaves of different position and differentseedling age were cultured in MS mediumsupplemented with 3mg/L 2,4-D and 0.5mg/L ABA.The M.I.peak of 49—72hseedlings appeared at 14∶00 only,andthose of 121—144h seedlings was fl-uctuately.The M.I.of 48 and 120h se-edlings were higher than those of 72,96and 144h seedlings.The M.I.decreasedgradually along the longitudinal directionof the foliar base,up to zero at 10mmaway from the base.The percentage of ca-llus formation was maximum at 0—3mmbasal segments,The percentage of callusformation of leaf basal segments from 120hseedlings was higher than those from 96and 144h seedlings.更多还原
