单克隆抗体NP28—5B对日本血吸虫卵肉芽肿形成的调节作用
EFFECT OF MONOCLONAL ANTIBODY NP28-5B ON SCHISTOSOMA JAPONICUM EGG-GRANULOMA FORMATION
【摘要】 本研究首先应用针对中国大陆株日本血吸虫可溶性虫卵抗原(SEA)中140kD靶抗原的单克隆抗体NP28-5B(lgG1),经体内被动转移试验表明对虫卵致敏/虫卵攻击的小鼠肺肉芽肿模型中日本血吸虫虫卵肉芽肿形成具显著的抑制或调节作用(抑制率为53%,P<0.002),进一步提供证据表明抗体参与虫卵肉芽肿形成的调节网络,也将为日本血吸虫病抗病免疫和抗病疫苗研究提供重要的分子基础。作者对上述作用机理作了有限探讨。
【Abstract】 In this study, we first evaluated the abilities of monoclonal antibody (Mab) NP28-5B (IgG1), specific for 140 kD molecule of soluble egg antigen (SEA) of S. japonicum (Chinese mainland strain), to modulate pulmonary egg-granuloma formation. The results showed that the in vivo administration of the Mab to egg-sensitized/egg-challenged mice down-modulated or modulated pulmonary granuloma formation (53% suppression, P< 0. 002). These data further confirmed the antibody-driven nature of some immunoregulatory networks in murine schistosomiasis japonica, and extend these observations to include Mab, and then provided an important information of molecular foundation for the study of antidisease immunity and the development of antidisease vaccines. In this paper, the authors explored primarily the possible mechanisms involved in the down-modulation of routine lung egg-granuloma formation by NP28-5B.
