薄层光密度法测定仙茅中仙茅甙的含量
QUANTITATIVE DETERMINATION OF CURCULIGOSIDE IN RHIZOMES OF CURCULIGO ORCHIOIDES BY TLC-DENSITOMETRY
【摘要】 本文报道用薄层光密度法测定仙茅中活性成分仙茅甙的含量。所用吸附剂为硅胶GF254,展开剂为氯仿-甲醇(4:1),样品以甲醇为溶剂用索氏提取器提取8h,提取液浓缩定容于2ml。采用岛津CS-930型双波长薄层扫描仪测定,测定波长282nm,参比波长340nm,反射法锯齿扫描,SX=3。加样平均回收率98.48%。并用此法对购自5个不同地区的仙茅进行了仙茅甙含量的测定比较,方法简便易行,结果稳定。
【Abstract】 The active principle curculigoside was isolated from rhizomes of Curculigo orchicides and identified by mp, IE. UV and TLO compared with standard of curculigoside. The crude drug was extracted in Soxhlet extractor with methanol. The methanol solution was spotted on a silica gel GF254 plate with a thickness of 300/u,m. and developed with chloroform-methanol(4:1). The spot of curculigoside was scanned on a Shimadzu Scanner CS-930 at λs = 282 nm,λR = 840nm, SX = 3 sensitivity med., reflection mode, zigzag scanning and determined by spot area, the average recovery was 98.48 %. The five samples of crude drugs from Shanghai, Kaifeng, Kunming, Nanjing, Wuning were analyzed.
