大鼠黄体细胞的长期培养

【摘要】 本实验建立长期细胞培养条件,观察了黄体细胞在无血清M199培养液中的分泌机能。结果发现随着黄体细胞密度增加,孕酮分泌量相应增多,24小时内培养液中孕酮含量随时间延长而增加。黄体细胞对hCG、Forskolin和佛波酮(PMA)等因素的刺激反应良好。

【Abstract】 Ovaries obtained from immature female rats treated with PMSG-hCG were sliced and dispersed into isolated luteal cells preparation by incubatinc. with collagenase and DNase.The cells were plated in 24-well culture plates at 3x105 cells per 0.5 ml culture mediura(medium 199 containing 2% fetal calf serum) per well and preincubated for 24h in a CO2 incubator ( 5% CO237C ) .After medium was replaced with serum-free medium , the cells were treated in the presence or absence of hCG, Forskolin or PMA for additional 4 h.Medium progesterone concentration was determined by RIA. Results are as follows: 1.There was a steady increase in progesterone production with the rat luteal cells numbers increase. 2.The progesterone production by rat luteal cells reached maximum in culture 24h. 3.hCG had significantly stimulated effect on progesterone release by rat luteal cells in vitro in a dose-dependent manner.4.Forskolin,an adonylate cyclase activator and PMA,a protein kinase C activator had significantly induced effecf on the progesterone production by Rat luteal cells.That demonstrated that both adonylate cyclase-cAMP-Protein kinase A and phos-pholipase C-IP3/DG-protein kinase C pathway involved in action of the hCG.
