甜菜坏死黄脉病毒分离提纯及血清制备

【摘要】 在甜菜坏死黄脉病毒分离提纯研究中,我们获得了一个较好的提纯程序。即选用新鲜病根组织加pH8.5硼酸缓冲液,用绞肉机研磨,纱布过滤,加6%PEG沉淀后差速离心,调节病毒悬液至酸性使病毒易悬浮,并通过病毒等电点沉淀法进一步纯化病毒。超迷离心所得病毒制剂呈典型的病毒核蛋白紫外吸收曲线,最大吸收值在260nm,最小吸收值在250nm,OD260/OD280为1.09。用所得BNYVV提纯液制备抗血清,其效价为1:128。

【Abstract】 In isolation and purification study of BNYVV, we obtain a better purification process. Buffer solution of boric acid (pH 8.5) which is put into fresh infected rootlets is pulverized by meat grinder and then filtered by gauze. After PEG precipitation, virus suspended solution is regulated to aridity so as to make virus easy to suspend, and virus can be purified by virus equal electric point precipitation, Typical virus ultiaviolet ray absorption curve of nucleoprotein can be seen from virus preparations attained through ultracentrifugation. Maximum absorption value is 260nm, minimum250nm, 0.D260/0.D280 is 1.09. Anti-serum with BNVVV purified solution has a titer of 1 : 128.
