环磷酰胺诱导大鼠血淋巴细胞染色体畸变与钙调素含量的关系
STUDY ON THE RELATIONSHIP BETWEEN CHROMOSOME ABERRATIONS AND CALMODULIN CONTENT OF LYMPHOCYTES FROM CYCLOPHOSPHAMIDE EXPOSED RATS
【摘要】 用5,10,15,20,25,30 mg/kg 环磷酰胺(Cy)染毒 Wistar 大鼠后,通过体外培养外周血淋巴细胞评定Cy 的潜在遗传毒性。结果,随 Cy 剂量的增加,染色体畸变率增加而分裂指数下降。用 ELISA 测定同批大鼠淋巴细胞内钙调素(CaM)含量,发现5mg/kg 组含量显著高于时照组,随 Cy 增加,CaM 逐渐降低。30mg/kg 组CaM 为10.47ng/10~6细胞,极显著低于对照组(19.28ng/10~6细胞),此种 CaM 含量的特异性降低可能是 Cy 抑制 DNA 合成所致。CaM 含量与染色体畸变呈直线相关,相关系数为—0.9984,与有丝分裂指数呈正相关,表明CaM 对细胞的增殖起着重要作用。
【Abstract】 The potential genotoxicity of cyclophosphamide(Cy)was assessed by in vitro cultivation of peripherallymphocytes of rats administered with Cy at doses of 5,10,15,20,25 and 30 mg/kg.The results showed thatfrequency of chromosome aberration(CA)increased and mitotic indices(MI)decreased with increasing of doses of Cy,revealing that Cy was a potential mutagen.Calmodulin(CaM)content of lymphocytes from experimental rats wasdetermined by ELISA.It was found that CaM level was remarkably higher than that of control at dose of 5 mg/kg,decreasing gradually with increase of Cy to 10.47 ng/10~6 cells at 30 mg/kg,which was significantly lower than that ofthe control(19.28 ng/10~6 cells).The decrease in CaM content might be attributed to the inhibition of DNA synthesis.CaM level and rate of CA was linear correlation with regression coefficient of—0.998 4.Positive correlation betweenCaM content and MI strongly suggested that CaM played an important role in the multiplication of cells.
