苎麻疫霉卵孢子萌发研究

【摘要】 苎麻疫霉(Phytophthora boehmeriae Sawada.)菌株在利马豆培养基平板上形成的卵孢子,用KMnO4和 H2O2处理,在 S+L 培养基上诱导萌发(25℃).结果表明,用 KMnO4和 H2O2处理卵孢子均能刺激其萌发.在一定时期内卵孢子萌发率随卵孢子保存时间的增加而上升,保存35d 的卵孢子用0.3%H2O2处理2min 和保存45d 的卵孢子用0.1%KMnO4处理20min,萌发率最高,分别为80%和92%.卵孢子萌发时给予光照对萌发有明显刺激作用,卵孢子在萌发过程中的光照条件以黑光灯连续照射效果最好,日光灯连续照射效果较差.所测定的4种培养基中以 S+L 对卵孢子的萌发效果最好,其次为SA 和 WA.

【Abstract】 Oospores of Phytophthora boehmeriae were treated with KMnO4 and H2O2 respectively,andgerminated on S+L medium for 12 days.Results showed that the germination of oospores was stimulated bythe pretreatment of oospores with those two oxidizers.The treatment with KMnO4 and H2O2 also preventedthe germination of sporangia and recidual mycelial fragments.Germination rate increased with the age ofoospores,reaching about 80% at 35-day-old and 92% at 45-day-old when oospores treated with 0.3%H2O2 for 2min,and with 0.1% KMnO4 for 20min,respectively.Illumination was essential for the germi-nation of oospores on S+L medium.The germination rate reached more than 80% when oospores were ex-posed to light and only 0~6% when light omitted.The best illumination for oospore germination was blacklight(near UV.),and the best medium was S+L.
