自制稳定的新型荧光剂CGE(N)的可用性探讨

【摘要】 用CGE(N)衍生化甘氨酸,衍生化最佳条件为pH8;温度50±1℃,时间3~5小时。甘氨酸-CGE(N)的薄层荧光扫描峰面积与其量的回归相关系数均在0.99以上。检测下限为0.24μg(以甘氨酸计),线性范围在0.24~19.2μg之间,稳定时间为24小时。结果表明CGE(N)的可用性和方法的可靠性。

【Abstract】 A new stable fluoresentreagent, CGE (N),was designed and syn-thesized in our laboratory. This paper pre-sents the results of using it to detect acompound having aminogroup. Glycine as an example was derived byusing CGE(N), the best condition of deri-vatisation reaction was pH8, T 50±1℃,and time 3-5h. Spread agent was CHCl3:CH3OH: CH3COOC2H5: iceHAc (1:3:5:1).Rf of glycine-CGE (N) was 0.45, of whichfluorescent spectrum Em was 356nm (atEx=277nm). The correlation coefficient between thefluorescent scanning area and quantitywas over 0.99, the linear relation retainedfor 24 h, the detection limit 0.24μg, andthe linear range 0.24-19.2μg.The data re-vealed that stability, sensitivity and re-producibility were good. These indicate that CGE(N) is a finefluorescent reagent for compounds havingprimary amino-group and this method isreliable.
