用酶联免疫吸附法测定多抗甲素含量的探讨
Study on Determination of Polyactin A by ELISA
【摘要】 作者采用酶联免疫吸附测定(ELISA)夹心法及二步竞争法,测定多抗甲素的含量。经实验条件的优化,均采用微滴板上烘干包被兔抗体,结合多抗甲素后,分别加入辣根过氧化物酶标记的免抗体或该酶标记的多抗甲素,再显色测定。除包被一步外,均在4小时内完成测定,比常规补体结合法快速,包被的微滴板4℃储存3个月仍不影响使用。灵敏度:夹心法为0.64~7.4μg/孔;二步竞争法为0.6~1.2μg/孔。
【Abstract】 Sandwich ELISA and two-step competition ELISA were utilized todetermine polyactin A.After optimizationof the experimental conditions, microtiterplates were coated with the rabbit antibodysolution and evaporated to dryness at 37℃.The cated microtiter plates were incubatedfirst with the analyte, then with the HRP-labelled antibody (sandwich procedures),or with the HRP-labelled polyactin A(com-measured after coloration. The whole de-termination took less than 4 hours, ex-cluding the coating procedure, which isfaster than the routine complement fixa-tion method. The coated microtiter platescan be well after over three months ofstorage at 4℃. The sensitivity obtainedwas 0.64-7.4ng/well for the sandwichmethod or 0.6-1.2μg/well for the com-petiton method.
