叠氮化钠对豌豆诱变效应的初步研究
Mutagenic Effects of Sodium Azide in Pisum satium L
【摘要】 豌豆种子在蒸溜水中预浸24h 后,放入0.000,0.002、0.020、0.200、2.000 m molL-1叠氮化钠溶液处理24h,研究叠氮化钠对豌豆的诱变效应。试验所用的测定指标包括 M1代的发芽率、田间出苗率、萌发延滞时间、根长、染色体畸变率、过氧化物酶、淀粉酶和蔗糖转化酶活性及 M2代叶绿素缺失突变率.试验结果表明:(1)高浓度叠氮化钠溶液(≥0.020mmolL-1)降低种子萌发率和田间出苗率,抑制根的生长,延滞种子萌发,抑制酶活性随萌发过程的增高;(2)低浓度叠氮化钠溶液(0.020mmol L-1)处理刺激根的生长和酶活性的增高;(3)叠氮化钠有效诱发 M2代叶绿素缺失突变。所有突变体中,黄绿苗突变体发生频率最高.0.200 mmol L-1叠氮化钠溶液处理,叶绿素缺失突变频率高达50.53%;(4)叠氮化钠不诱发根尖染色体畸变。
【Abstract】 Seeds of pea(Pisum salivum L) were presoaked in distilled water for 24h and treated with 0、0.002、0.020、0.200、2.000 m mol L-1 sodium azide solutins to study mutagenic effects of sodium azide in pea.Criteria used in the experiment included germination、field seedling、root length、chromosomal aberration frequency,activities of peroxidase、amylase and in- vertase in M1 generation as well as chlorophyll—deficient mutation frequency in M2 germina- tion.The experimental results showed:(1)the sodium azide treatments of higher concentration(≥ 0.020 m mol L-1) decreased germination and field seedling,inhibited root growth,delayed germi- nation and inhibited the increases of enzymes along with germination course;(2)the sodium azide treatment of lower concentration(0.0020 m mol L-1) stimulated rooth growth and the increases of enzymes;(3)sodium azide proved effective for inducing chlorophyll deficient mutations in M2 generation.Among all chlorophyll—deficient mutents,the frequency xantha—viridis was the high- est.The frequency of chlorophyll—deficient mutations induced with 0.200 m mol L-1 azide solu- tion was as high as 50.53%;(4)sodium azide did not induce any chromosomal aberration of root tip.
