球孢白僵菌017壳聚糖酶的研究I酶制剂的基本特征

【摘要】 球孢白僵菌017突变菌株(Beauveria bassiana 017)在CTN培养基(含0.2~1%壳聚糖,0.075%KH2PO4,0.05%MgSO4,0.05%NaCl,0.001%FeSO4,0.3%KNO3,0,1%牛肉膏,pH5.0)中,20~30℃,200r·p·m振荡培养7天,可以分泌大量的胞外壳聚糖酶,对底物水解的最适温度为45~50℃,最适pH值为3.8~4.0。Km值和Vmax分别为2.17×10-6M和0.45μmol/min,60℃保温1小时,剩余酶活性为10%,Ca2+,Mn2+和Mg2+对酶有激活作用,Cu2+和Fe3+对酶有部分抑制作用,Hg2+和Ag+则强烈抑制酶的活性。

【Abstract】 Beauveria bassiana 017 incubated in CTN medium (contained 0.2-1 % Chitosan, 0.075% KH2PO4, 0.05% MgSO4, 0.05%NaCl, 0.001%FeSO4, 0.3 % KNO3 and 0.1% beef extract, pH 5.0 at 20-30℃ and 200rpm for seven days can secret Chitosanase quantitatively. The optimum pH and temperature,the Km value and Vmax for Chitosanase were 3.8-4.0, 45-50℃, 2.17× 10-6M and 0.45μmol/min,repectively. Chitosanase lost 90% of its activity at 60℃ for 1 hour. This enzyme was activated by Mn2+,Mg2+ or Ca2+,while strongly inhibited by Hg2+ or Ag+,and partly inhibited by Cu2+ or Fe3+.
