二株抗人原发性肝癌AFP—McAb杂交瘤的建立
Establishment of two Hybridoma Cell Strains Secreting Monoclonal Antibodies Against Human Primary Hepatocellular Cancer AFP
【摘要】 本文报道从GHC—1及GHC—3分泌的AFP,经过常规方法的改进,获得AFP—McAb。以5pg/kg AFP免疫后的BAL B/C小鼠脾细胞与SP2/0小鼠骨髓瘤细胞融合,采用ELISA—Ⅰ、DIBA、RPHA—Ⅰ和杂交瘤细胞免疫荧光法,筛建了二株可分泌抗人原发性肝癌的AFP—McAb杂交瘤,定名为GAH1及GAH2。 RPHA—Ⅰ测试表明,GAH1及GAH2分泌的McAb,可抑制原发性肝细胞癌病人的腹水和血清,但对脐带血不能完全抑制。
【Abstract】 AFP-McAb was obtained from AFP secreted from GHC-1 and GHC-3 by the modified conventional methods. After the fusion between the splenic cells from BAL B/C mice immunized with 5μg/kg AFP and mouse myeloma cells SP 2/0, then applying ELISA-I,DIBA, RPHA-I and immunoflourescence staining, the hybridoma cell strains were screened. We succeeded in establishment of two hybridoma cell strains capable secreting anti-human primary hepatocellular cancer AFP-MeAb. These two cell strains were named GAH1 and GAH2. RPHA-I study showed that the MeAb secreted by GAH1 and GAH2 reacted inhibited with sample of human primary hepatocellular cancer, such as aseites and serum of the patients, but incomplete inhibited with cord blood.
