可溶性人SCF在大肠杆菌中高效表达
High Expression of Soluble Forms of Human Stem Cell Factor in Escheria Coli
【摘要】 干细胞因子(SCF)是参与机体生长发育过程中的一种多功能细胞因子,该因子在造血早期发挥重要作用。本研究利用PCR技术克隆分泌型可溶性人SCF cDNA,并将其转入大肠杆菌中进行表达,结果显示,表达产物占菌体总蛋白20%以上,主要以包涵体形式存在,经变性和复性后,证明具有SCF的生物学活性。
【Abstract】 Stem Cell Factor (SCF) is a multiple functional growth factor involved in human body development.It plays an important role in the early stages of hematopoiesis. In present research, PCR technique was usedto clone the cDNA for soluble form of human SCF, then the cDNA was transferred into Escheria coli forexpression. The result showed that expressed proteins are about 20KD in size and account of more than20% of total bacterial proteins. They exist in insoluble form of inclusion bodies. After a series of denatureand recovery treatment, the expressed protein was confirmed to have biological activity of SCF.
