牛体外受精卵的冷冻保存及解冻后的胚胎移植

【摘要】 将来自体外受精的牛桑椹胚和囊胚分别以乙二醇和甘油为冷冻保护剂,利用常规方法冷冻保存,观察了其冷冻效果。另外,将一批冷冻-解冻胚以直接移植法(胚胎不去除防冻剂移植)和常规移植法(胚胎去除防冻剂后移植)移植给受体母牛,观察了受胎情况。结果表明,乙二醇对于牛体外受精卵的冷冻保存效果明显优于甘油;采用直接移植法对冷冻保存后的牛体外受精卵移植后,同样能获得受胎的结果。

【Abstract】 The morulae and blastocysts derived from bovine IVM and IVF embryos were frozen by using ethylene glycol (EG) and glycerol (Gly). The effect of sucrose in EG treatment on freezing of embryos was examined. Some frozen-thawed bovine IVF embryos were transferred to recipients nonsurgically by adopting direct or conventional transfer methods, with the former keeping cryoprotectant while the latter removing cryoprotectant. The results showed that the effect of EG on the freezing of bovine IVF embryos was significantly better than the effect of Gly. The survival rates of embryos thawed after culturing for 48 hours were 63.8% and ,12.3% (P<0.05) respectively. Sucrose added in EG treatment was not beneficial to the freezing of the embryos. The pregnancies could also be realized by direct transfer method after freezing of bovine IVF embryos. The conception rates were 41.7% in direct and 30.0% in conventional transfer method respectively.
