抗癌抗生素C1027与单克隆抗体偶联物抗胃癌作用的实验研究
Activity of Immunoconjugate Composed of Antitumor Antibiotic C1027 and Monoclonal Antibody Directed against Gastric Cancer
【摘要】 抗癌抗生素C1027对体外培养的癌细胞有很强的杀伤作用。抗人胃癌单克隆抗体3H11与C1027连接,偶联物保留抗体对靶细胞的反应性。克隆形成测定表明,偶联物对人胃癌BGC-823细胞有很强的特异性杀伤作用,IC50为2.0×10-17mol/L。裸鼠体内试验表明,偶联物对人胃癌BGC-823移植瘤的生长有显著抑制作用,在可耐受剂量(0.1mg/kg)的肿瘤抑制率为83%。
【Abstract】 Antitumor antibiotic C1027, which is produced with a streptomyces strain, shows highly potent cytotoxicity to cultured cancer cells. C1027 was conjugated to 3Hll, a monoclonal antibody directed against gastric cancer cells, by using SPDP as linking agent and the resulting immunoconjugate retained antibody reactivity. As shown by clonogenic assay, the 3Hll-C1027 conjugate is highly potent and selective cytotoxicity to gastric cancer BGC-823 cells with IC50 value of 2.0.×10-17mol/L. It also exerts marked suppression on the growth of gastric cancer BGC-823 xenograft in nude mice with 83% inhibition at the tolerable dose of 0.1mg/kg.
