人βE-珠蛋白基因转基因鼠的建立
Establishment of Transgenic Mouse Lines of Human βE-globin Gene
【摘要】 利用显微注射法制备了人βE-珠蛋白基因的转基因小鼠系,其外源基因(βE)能通过生殖细胞稳定地传递给子代,具有典型转基因鼠特征。证实人βE-珠蛋白基因在转基因鼠中的高水平表达需有红系增强子结构(5′HS2)存在。单一位点整合过多拷贝(16拷贝)人5′HS2βE-基因的平均表达水平明显低于较少拷贝(2拷贝)整合的表达水平。
【Abstract】 The human βE-globin genes with or without erthroid enhancer 5′HS2 are microinjected into the firtilized mouse lines. The transgenic mouse line bearing 5′HS2 βE-globin gene has such typical characteristics that the transgene could be stably transmitted into its offspings through the germ line. It is proved that the high level expression of the human βE-globin gene in transgenic mice needs the presence of 5′HS2. The expression level per gene copy of 5′HS2 βE is significantly reduced when too many copies integrated in a single site in comparison with that of mouse bearing fewer copies of 5′HS2 βE.
