金属硫蛋白的扫描隧道显微镜观察与分析
Structure of Metallothionein Imaged by Scanning tunneling Microscopy
【摘要】 将溶于双蒸水的金属硫蛋白(MT)分子滴在石墨表面,经加热展层,再自然干燥。用DS-89型恒流模式STM进行观察,我们成功地获得了稳定的MT分子图象,并可观察到局部区域的肽链走向,为蛋白质高级结构的研究提供了一种新的补充方法。
【Abstract】 Metallothionein(MT)dissolved in double-distilled water was dropped onto graphite and heated to 80℃ for 20 seconds,then dried in the air.Through the DS-89 constant-current mode STM,we not only succeeded in obtaining images of MT molecules,but also found the peptide chain at some parts.Our work gives a new method to the research of the three-dimensional conformation,though it has not been matured yet.
