狗脑抗吗啡镇痛肽“60a”的分离纯化及部分一级结构
The Isolation, Purification, Sequince of an Antimorphine Peptide(60a) from Dog Brain
【摘要】 本实验室从正常狗脑中又分离获得一个暂称之为“60a”的肽。冻干粉经SephadexG-50和羧甲基纤维素CM22柱层析,再经反相高效液相层析纯化得最后样品,经SDS电泳和等电聚焦电泳鉴定均为一条带,分子量约8000,等电点在pH7.8左右;N-末端氨基酸为Met;自N-末端的部分氨基酸序列为:H2N-Met-Leu-Ser-Pro-Ala-Asp-Leu-Thr-Asp-Ile-Leu-Phe-Thr-His-Asp-Leu-Ile-Gly-Gly-His-Phe-Arg-Pro-Ala-Lys-Tyr-Lys-Lys-His-Pro-Val-His-Trp-Phe-Val-Ile-Trp-Trp-Pro-……。
【Abstract】 A pure peptide named 60a from dog brain was obtained. The whole dog brain was homogenized and extracted with 0.1mol/L acetic acid. The supernatant was lyophilized and the residue was dissolved in 0.1mol/L acetic acid and was allowed to go through Sephadex G-50 column (twice), CM22 cellulose. The sample obtained was applied to a rp-HPLC with C18 column. The sample was named 60a peptide, and showed only one band on SDS-polyacrylamide gel electrophoresis or isoelectric focusing. Its isoelectric point was around pH 7.8 and M. W. was about 8000 dalton. The sequince of the N-terminal 46 amino acid residues has determined. Its N-terminal amino acid was Met. The 60a peptide showed a significant anti-morphine activity when intravetricularly injected to the mice.
