大蒜素等对MNNG及环磷酰胺诱发V79细胞微核形成的影响

【摘要】 为了弥补原核生物短期试验揭示诱变抑制剂之不足,建立了体外培养哺乳类V79细胞微核抑制试验检测诱变抑制剂,并对该试验作了区分去诱变剂和抗诱变剂设计,辅以受试物对细胞生长影响,以揭示真正的诱变抑制剂。实验证明,大蒜素0.06mg/L、0.6mg/L能降低MNNG 0.1mg/L和环磷酰胺2mg/L诱发V79细胞微核形成率(P<0.05),但它对细胞增殖亦有抑制作用。白术水提取液1.25×103mg/L、5×103mg/L在代谢活化系统存在情况下,可抑制环磷酰胺2mg/L诱发V79细胞微核的形成(P<0.05),推测它可能对代谢活化酶系有揶制作用,有待进一步实验证实。

【Abstract】 An assay based on the inhibition of micronucleus formation in mammalian V7?cells in vitro was established, as an insubstituted short-term test for screening themutation inhibitor, in order to overcome the deficiency of prokaryote tests. In thisassay, a design for distinguishing the desmutagen from antimutagen was made. Alsoincorporated in this assay was the supplementary test for observing the effect of thetest substance on the cell growth, in order to discover the actual inhibitor ofmutagenesis by micronucleus inhibition assay. The result showed that the diallyl trisulfide, a biological effective constituent ofgarlic (Allium sativum L.), at the concentration of 0.06 and 0.6 mg/L can decreaseV79 cell micronucleus formation frequency induced by 0.1 mg/L MNNG and 2 mg/LCP (P<0.05) while it also suppress cell proliferation markedly at a concentration of0.06 mg/L. The result also showed that V79 cell micronucleus formation was also inhibited bythe aqueous extract of Atractylodes macrocephala at the concentration equivalent to1.25×10~3, 5×10~3 ug raw material/ml induced by 2 mg/L CP at the presence of meta-bolic activation (P<0.05), but no inhibitory effect was observed on those induced byMNNG directly. It is suggested that Atractylodes macrocephala extract may act as aninhibitor of metabolic activation enzyme, this requires further experimentalvalidation.
