一种人体雌激素受体和孕酮受体的微量检测法

【摘要】 本文重点介绍一种琼脂糖凝胶等电聚焦电流(IEF法)对乳腺癌组织雌激素受体(ER)及孕酮受体(PR)的测定方法。鉴于该检测法所需组织样品量小,不仅可对手术切除组织进行测定,还能应用于活检标本。本文通过39例人体乳腺癌标本及1株MCF-7乳腺癌细胞株的检测,采用IEF法测定ER、PR,其所需组织量仅50mg。此法操作简便,实验重复性好,并能同时进行多种激素受体的测定。

【Abstract】 We introduce a method of detecting the Estrogen Recoptor and ProgesteroneReceptor in human breast cancer by isoelectric focusing in flat beds of agarose gel.Owing to the very small amount of tissue(50mg)required in this method,it can be usedto detect the ER the PR content not only in mastectomy tissues,but also in the needlebiopsies. We detected the ER and PR content on thirty-nine cases of breast cancer andone MCF-7 breast cancer line by this method with the results aualogous to that of DCCmethod. The technique procedure is very simple and well repeatable and allows severalkinds of hormonal receptors to be determined at the same time.
