应用高压液相色谱测定苏云金杆菌发酵液中的苏云金素

【摘要】 通过正交试验将测定苏云金素所用的HPLC流动相确定为:0.075M磷酸二氢钾水溶液,加1%甲醇并用磷酸调pH至2.5。所选定的色谱条件可保证苏云金素的分离度为100%。苏云金素浓度从0.6μg/ml至900μg/ml,其峰面积与素的含量之间呈很好的直线关系。苏云金素的最低检出量为2.25×10-3ng。用发酵样品进行测定,不同浓度素的峰面积对数与生测的试虫死亡机率值之间的相关系数为0.998。以HPLC法和昆虫生测法同时测定结果各重复间的变异系数(C.V.)为0.2~1.5%,测出浓度与实际浓度的平均不符合率为2.4%;昆虫生测法的测定结果各重复间的变异系数为14.6~16.2%,测出浓度与实际浓度的平均不符合率为17.0%。用这两种方法同时测定了3个苏云金素发酵样品中素的浓度,两种方法所得结果的平均符合率为86.84%。

【Abstract】 A mobile phase for determining thuringiensin in the ferment broth ot Bacillus thuringiensis by high-performance liquid chromotography (HPLC) was selected by factor tests. The mobile phase was 0.075 M KH2PO4-water solution plus 1% methanol, adjusted to a pH of 2.5 with phosphoric acid. Under this condition, the separative efficiency of thuringiensin was 100%. The content fo thuringiensin was in a linear correlation with the peak area over a concentration range of 0.6 to 900 ug/ml. The minimum detectable amount of thuringiensin was 2.25×10-3n. Compared with bioassay of thuringiensin with insects, the result of HPLC was within 2.4% of true value, while that of bioassay was within 17% of true value. Results of HPLC and bioassay coincided at a frequency of 86.8%.
