苜蓿体细胞胚发生与成苗研究 Ⅲ.脱落酸处理提高体细胞胚的质量

【摘要】 在液体培养基中诱导产生的苜蓿体细胞胚,诱导第12天时生活力较高。超过12天后,体细胞胚由绿色向褐色转变,体细胞胚肥大肿胀,最后死亡。若将誘导12天产生的体细胞胚转移到成分相同的固体培养基上继续培养,则体胚进一步发育和转化成小苗。在固体培养基中加入脱落酸(ABA),可以抑制体细胞胚萌发,促进体细胞胚健康度提高,获得高质量的成熟休细胞胚。在固体培养基诱导产生的体细胞胚健康度比液体培养基中誘导的高得多。

【Abstract】 The viability of alfalfa somatic embryos induced in a liquid medium was high on the 12th day after culture. After that time, the somatic embryos swelled and bacame brown, finaly lost their viability. When the somatic embryos induced in the liquid medium were further cultured on a agar medium, the embryos continued to develop into plantlet. The treatment with ABA prevented precocious germination and increased the quality of somatic embryos. Somatic embryos induced in agar medium began to germinate precociously on the 14th day after culture. The somatic embryos of higher vigor were obtained when they were induced in the agar medium with ABA.
