c-myc反义RNA对NIH/3T3、人胃癌细胞生长及生物大分子合成的影响
EFFECTS OF C MYC ANTISENSE RNA ON CELL GROWTH AND BIOLOGICAL MACROMOLECULE BIOSYNTHESIS OF NIH/3T3 CELLS AND HUMAN GASTRIC CANCER CELL LINES
【摘要】 采用c-myc反义RNA表达质粒与pSV2neo质粒经电击穿孔技术共转染了NIH/3T3细胞和人胃癌823细胞,经适当浓度G418筛选后,获得其相应的转染细胞株。实验表明,该转染细胞在一定浓度Cd2+诱导c-myc反义RNA表达后,细胞形态有所改变,生长速度受到抑制,DNA、RNA及蛋白质生物合成均有不同程度的变化。
【Abstract】 NIH/3T3 cells and human gastric cancer cell line 823 were cotransfected with c-myc antisense RNA expression plasmid and pSV2neo plasmid by electroporation method. Transfected cell lines selected by G418 were obtained. The induction in transfected cell of c-myc antisense RNA expression by Cd2+ with certain concentration resulted in alteration of cell morphology, inhibition of cell growth and changes in DNA, RNA and protein biosynthesis.
